Polyclonal antibodies (pAbs) are a mixture of antibodies created and secreted by B Cells. These antibodies are produced in reaction to different epitopes on an antigen’s surface. The most commonly used species for this type of polyclonal production are rabbit and sheep.
pAbs are relatively quick and easy to produce, and are more importantly, cost-effective as compared to Monoclonal Antibody Production. Our services are outlined below.
Option A: Custom polyclonal antibody production using protein antigen
- Protein Approval: The customer must provide their own protein provided we are happy with the purity and buffer components.
- Immunisation: We use a standard 90-day immunisation protocol consisting of four immunisations in your choice of host animal (see Table 1).
- Purification (optional): We purify up to 20ml of the final sera using the Protein A.
Option B: Custom polyclonal antibody production using peptide antigen(s)
- Peptide Design: We select peptides based on the primary and secondary structure of the native protein, and the cross-reactivity with other proteins within the same species.
- Peptide Synthesis: Our peptides for immunisation are always >80% pure. We synthesise 18mg in total of the chosen peptide, 2mg of this is aliquoted separately for the customer to use as they wish.
- Conjugation to Carrier Proteins: We conjugate the peptide(s) to KLH and BSA for immunisation and testing respectively.
- Immunisation: We use a standard 90-day immunisation protocol consisting of four immunisations in your choice of host animal (see Table 1).
- Affinity Purification (optional): We purify up to 20ml of the final sera using the peptide(s) used for immunisation.
Option C: Custom polyclonal antibody production using a PTM-specific peptide antigen
- Peptide Design: We select the best possible peptide sequence to include the region of the desired PTM.
- Peptide Synthesis: Our peptides for immunisation are always >80% pure.We synthesise 18mg in total of the chosen peptide, 2mg of this is aliquoted separately for the customer to use as they wish. We will also synthesise the required peptide without the PTM in place in order to deplete non-specific antibodies at the purification step.
- Conjugation to Carrier Proteins: We conjugate the peptide(s) to KLH and BSA for immunisation and testing respectively.
- Immunisation: We use a standard 90-day immunisation protocol consisting of four immunisations of the modified peptide in your choice of host animal (see Table 1).
- Affinity Purification (optional): We purify up to 20ml of the final sera using the peptide(s) used for immunisation. We will purify the sera using the non-modified peptide in order to deplete non-specific antibodies from the sera. We will then purify the same sera using the modified peptide used for immunisation.
